Induction of Genetic Damage in Peripheral Erythrocytes of the Fish, Anabas testudineus Exposed to Sublethal Concentration of Acid Orange 7

Abstract


Acid orange 7, a synthetic organic compound, generally considered as pollutant, released from the process of chemical textile finishing process was exposed to the freshwater fish Anabas testudineus at sublethal concentration i.e., 0.27 gl-1 for 24, 72 and 96 h, maintaining ten animals per group along with the control. The median lethal concentration of acid orange 7 determined by Probit analysis was 2.7 gl-1 concentration. Sublethal exposure to the azo dye caused genotoxicity as evidenced by the formation of micronucleus along with other nuclear abnormalities such as irregular, notched and blebbed nucleus. Exposure to acid orange 7 also showed alterations in the comet parameters such as comet area, head area and tail area on the erythrocytes of the fish, Anabas testudineus. The other comet parameters such as tail moment and olive moment also increased in time-dependent manner representing the DNA damaging potential of acid orange 7. The study used simple, cost-efficient, fast and reliable methods for the detection of genetic damage induced by acid orange, and thus provides great value and suitability as a bio-indicator of genotoxic stress.
