Analysis of Genetic Basis for Morphological Variations in Normal and Tissue Cultured Plants of Andrographis paniculata in Association with or Without Arbuscular Mycorrhizal Fungi using AFLP Markers

Abstract


Andrographis paniculata Nees, a member of Acanthaceae, is one of the important medicinal herbs. The active principle, Andrographolide esters has a paramount importance in pharmaceutics. Morphological variants have been reported both in natural populations collected from various geographical areas and micropropagated plants raised through tissue culture that need to be analyzed with proper molecular tools. In order to facilitate reasoned scientific decisions on its management, conservation and prepare for a selective breeding programme, genetic analysis of normal and tissue cultured plants with or without arbuscular mycorrhizal fungi association was performed using amplified fragment length polymorphism markers. Out of 15 primer combinations of EcoR-I/Mse-I and Pst-I/Mse-I, six combinations of Pst-I/Mse-I showed reproducible and well-resolved bands. Six primer combinations scored 641 bands, out of which 495 were polymorphic revealing 77.22% of polymorphism. Amongst six samples analysed, tissue cultured plants showed a highest of 90.50% of polymorphism and least was found in normal plants treated with Glomus mosseae with 62.80%. Numerical taxonomy and multivariate analysis version 2.02e software (NTSYSpc) and unweighted pair group method with arithmetic mean (UPGMA) were used to generate similarity co-efficient and dendrogram. The dendrogram generated based on the similarity matrix has clustered the samples according to their parentage and treatment.
