Comparative Study on the Effect of Growth Regulators on The Development and Multiplication of in vitro Germinated Orchids of Bangladesh

Abstract
‘In vitro’ culture response of different plant growth regulators was assessed in two indigenous medicinal orchid species namely, Acampe papillosa (Lindl.) Lindl. and Aerides odoratum Lour. orchid for optimum callus induction and plantlet regeneration. Leaf and nodal segments of in vitro grown seedlings of the both species were cultured on MS and PM (Phytamax) media supplemented with different concentration and combinations of plant growth regulators of Auxin and Cytokinins. The nodal segments of both species gave positive response producing different types of calli as well as multiple shoot buds depending on nutritional stimulus. Leaf segments of both the species failed to response to any of the PGR combinations. In subsequent subcultures, the callus tissues underwent differentiation via PLB formation on a broad spectrum of PGR supplemented media. Strong and stout root system was induced in half strength MS medium supplemented with 0.5 mgl-1 IAA. The well-root plantlets were transferred to pots containing a potting mixture composed of saw dust, coconut coir, humus and coal pieces at 1:1:1:2 (w/w) with survival in outside natural environment through successive phases of acclimatization.
