Isolation and Screening of Some Protease Producing Bacteria to Treat Paper and Pulp Effluent

Abstract


Industrial pollution is one of the most alarming threats to the environment and paper and pulp mill is one of the prime contributors to it. Paper and pulp mill effluent affects biotic and abiotic components of the environment. Different methods are being employed for the treatment, but the biological method is found to be more efficient in terms of colour removal and for reduction in the value of BOD, COD, TDS and TSS. Hence, the present study was designed to isolate and to screen some protease producing microorganisms which shows biodegradation potentials to treat paper and pulp effluent. In this study, about 25 isolates were isolated from the different effluent samples collected from the paper and pulp industries in Vapi, Gujarat. They were studied for the decolourisation and physicochemical parameters before and after decolourisation studies. Results showed that the isolate S2and S6 showed the highest decolourisation followed by L2, L6,G2and G8. Further, theses six isolates were screened for selected enzyme production, i.e., Amylase, Laccase, Protease and Pectinase. S6showed the highest zone diameter for protease, i.e., 2.0 mm. Among these six isolates, S6 showed the best Protease production, i.e., 40 U/ml on the fourth day of incubation.
