

Response of Cytokinins on In Vitro Shoot Multiplication of Rose cv. Frisco

Abstract
This research was performed to investigate the response of cytokinins on in vitro shoot multiplication of rose cv. ‘Frisco’. The nodal segments were cultured on MS medium containing 2.0 and 10.0 mg L-1 of 6-benzylaminopurine (BA), 2.0 mg L-1 and 10.0 mg L-1 of thidiazuron (TDZ) and, 2.1 mg L-1 and 10.5 mg L-1 of meta-topolin (MT). Inclusion of BA and TDZ in the culture medium promoted multiplication of shoots and a maximum number of shoots (3.75) was recorded both from 10.0 mg L-1 of BA and TDZ. Although highest number of shoots was obtained from the medium with 10.0 mg L-1 BA but incidence of hyperhydricity symptoms was appeared in the new micro-shoots. MT with 2.1 mg L-1 found to be effective for vegetative growth by securing highest elongation of shoot (2.59 cm) and developing maximum number of leaves per shoot (8.50). Among the three investigated cytokinins, TDZ at 10.0 mg L-1 is the optimal plant growth regulator for in vitro shoot multiplication of rose cv. ‘Frisco’.
