In vitro Propagation of Geranium (Pelargonium Graveolens L.) from Nodal Culture

Abstract
The present study was initiated to optimize in vitro propagation protocol from nodal culture of geranium. Three surface sterilization treatments were tested, and treatment three-surface sterilization (treatment of explants with 1.5%(w/v) fungicide (Kocide 101) mixed with one drop of soup solution for 1 min, 70% ethanol for 1 min and with 1% NaClO for 3 min and again with 70% ethanol for 1 min was used throughout the experiment with minimum contamination and low percentages of dead nodal explants. The nodes were cultured on MS-medium supplemented with different combinations and concentrations of BAP and IAA and on hormone-free MS-medium. Data were collected and subjected to analysis of variance (ANOVA) and significant means were separated using Duncan multiple range test at α = 5% significant level. The maximum percentage of node-initiated shoots (93.33%) were observed on hormone-free MS-medium after four weeks of nodal inoculation. But the lowest percentage (33.33%) of nodes initiated shoots were recorded from MS-medium supplemented with 2.0 mg/L BAP + 2.0 mg/L IAA. MS-medium was also used for multiplication of shoot from nodal culture. The hormone-free MS-medium recorded the highest mean number of shoots/explants (11.50 ± 2.64) and the highest mean length of shoot (2.06 ± 0.04 cm) at the end of the fourth week, while the lowest mean number of shoots/explants (1.17 ± 0.29) and shoot length (0.50 ± 0.10 cm) were recorded on MS-medium supplemented with a combination of 5 mg/L BAP and 1 mg/L NAA. The best rooting was observed on the hormone-free full-strength MS-medium with mean number of main roots (22.60 ± 1.14) and mean root length (6.34 ± 0.11 cm). The lowest mean number of main roots (3.80 ± 0.83) and mean root length (1.18 ± 0.21 cm) were recorded on half strength MS-medium containing 1 mg/L IBA after four weeks of shoot sub-culture. Finally, 77.78% of plants survived at the end of primary acclimatization and 75% plants survived at the end of secondary acclimatization process. The hormone-free MS-medium was the best for shoot initiation, multiplication and rooting.
